One way the Bailey Lab examines protein-nucleic acid interactions is to determine the structure of the molecules involved. In the past, we’ve used x-ray crystallography to solve these structures, but Cryo-EM has a number of advantages – for example, it’s useful for larger structures, such as CRISPR complexes made up of multiple proteins, and unlike x-ray crystallography it doesn’t require all the special preparation to crystallize the molecules.
Now that the cryo-EM is up and running, we’re looking forward to using it to further our investigations of protein-nucleic acid interactions at a functional and mechanistic level.
